Scale & Synthesis Strategies
We manufacture peptides up to 50 amino acids in length and in quantities that can range from gram to multi-10 kg quantities. Our robust, economical manufacturing processes will help optimize the costs of development.
Several synthesis strategies are available. The choice will be based on the particular sequence, as well as your specific project needs.
Large-Scale Liquid (Solution) Phase Peptide Synthesis (LPPS)
LPPS is the technology of choice for many peptides, particularly shorter peptides or structures that prevent attachment to a resin matrix. We currently provide more than 11,000 L of reactor volume for LPPS projects.
Large-Scale Solid Phase Peptide Synthesis (SPPS)
SPPS is well-suited to manufacturing peptide APIs, particularly longer, more complex sequences. PolyPeptide Laboratories has extensive experience in both Fmoc and t-Boc protection strategies and we will work together to identify the optimal synthetic approach, depending on peptide sequence length and your product requirements. We maintain SPPS reactors from 10 L to 300 L in volume, and can manufacture up to 200 kg quantities of peptides with complex sequences using SPPS technology.
Large-Scale Hybrid Fragment Synthesis
In hybrid fragment synthesis, small peptide fragments are produced using SPPS and then joined in solution to form longer peptides. For some longer peptides this approach offers a more efficient process than "straight-through" SPPS. Because "straight-through" Fmoc-based SPPS and hybrid fragment strategies usually use the same Fmoc-amino acid derivatives, switching synthetic strategy usually has a minimal impact on impurity profiles.
Large-Scale Purification
From development to manufacturing, purification is key to any project's success. PolyPeptide Laboratories has the technology, equipment and expertise to attain product purity at any scale, from small- to high-volume, including:
- Preparative reversed-phase HPLC
- Ion exchange chromatography
- Size exclusion chromatography
- Columns up to 60 cm internal diameter
Large-Scale Isolation
Peptide APIs are typically isolated by lyophilization. PolyPeptide Laboratories is equipped to process multi-kilogram lots of product using multiple flask lyophilizers as well as tray lyophilizers with up to 200 kg ice capacity. We can also explore alternatives such as precipitation and spray drying, which can be scaled up to raise throughput in large-scale processes.
