Expanding the Reach of Sustainable Solid-Phase Peptide Synthesis

Alloc protecting group expanded the peptide synthesis toolbox. Its removal can sometimes be seen as an issue: toxic solvents, air-sensitive Pd0) reagents, challenging upscaling. Not anymore

A new method eliminates metal catalysts and hazardous reagents. It uses iodine/water in PolarClean and EtOAc to enable:

  • On-resin Alloc removal
  • One-pot removal + coupling
  • Full compatibility with Fmoc and Alloc amino acids
  • Seamless use with long peptides (achieved with a 39 amino acid peptide)
  • Successful upscaling to 10 g
  • TFA-free cleavage
